Gene Bio Solution ALK-1 Antibody. Gene bio solution’s recombinant rabbit antibody recognizes ALK-1 (Anaplastic Lymphoma Kinase), a receptor tyrosine kinase that belongs to the insulin receptor superfamily. ALK-1 plays an important role in the development of the brain and exerts its effects on specific neurons in the nervous system. The ALK-1 gene has been found to be rearranged, mutated, or amplified in a series of tumors including anaplastic large cell lymphomas, neuroblastoma, and non-small cell lung cancer. The chromosomal rearrangements are the most common genetic alterations in the ALK-1 gene, which result in creation of multiple fusion genes in tumorigenesis, including ALK-1/EML4, ALK-1/RANBP2, ALK-1/ATIC, ALK-1/TFG, ALK-1/NPM1, ALK-1/SQSTM1, ALK-1/KIF5B, ALK-1/CLTC, ALK-1/TPM4, and ALK-1/MSN. The specificity of the ALK-1 antibody (clone 1A4) was established on known ALK-1 positive and negative non-small cell carcinoma (NSCLC). The antibody to ALK-1 presented no staining on formalin fixed ALK-1 negative NSCLC tissue and positive staining on ALK-1 positive NSCLC tissue using immunohistochemistry.

ALK-1 is typically expressed at low levels in regions of the developing central and peripheral nervous system. ALK-1 may be activated in cancer through multiple mechanisms. The most common mechanism is through the formation of a fusion protein from chromosomal translocations, as in the case of anaplastic large cell lymphoma (ALCL) and inflammatory myofibroblastic tumors. ALK-1 may also be amplified through mutation, as in neuroblastomas. Various solid tumors, such as non-small cell lung carcinoma (NSCLC) and brain cancers were also found to aberrantly express ALK-1. ALK-1 staining is present within both the nucleus and cytoplasm and is positive in about 60% of ALCL. ALK-1 protein expression by tumor cells is an independent prognostic factor that predicts a favorable outcome.
