Goats are immunized with purified human C-4 to produce antiserum against it. The antiserum is then treated to obtain the gamma globulin fraction and reacted with fluorescein isothiocyanate. After removing the excess dye through dialysis, the conjugated globulin is further purified on DEAE cellulose to eliminate any protein that is under or over labeled. The total protein content is determined using the Biuret reaction, while the concentration of fluorescein is compared to a fluorescein diacetate standard. To preserve the product, sodium azide (0.1% w/v) is added. When antigen-antibody complexes form and complement fixation occurs, Human C-4 (B1E) is converted into the inactive products C4a and C4b. The antiserum used in the production of this conjugate reacts with both intact C-4 (B1E) and C4b. Therefore, a positive fluorescence with this conjugate does not allow differentiation between active C-4 and its breakdown product.
